Optimization of Culture Conditions Using the Method of Response Surface Methodology for Chitosanase Production by Enterobacter sp. YT21

Abstract
Optimization of culture conditions were conducted for chitosanase production by Enterobacter sp. YT21. Firstly Culture temperature, pH and inoculum volume were optimized by one-factor-at-a-time experiments, and then Box-Behnken design and response surface methodology were employed for further optimization of the chitosanase production. The results indicated that the optimal culture conditions were 31.74℃, pH 6.5 and 7.26% (V/V) inoculum, and the highest chitosanase activity obtained was 13.59 U/mL. Error of the actual value compared with the predicted one was only 0.43%, so Enterobacter sp. YT21 was proved to be a suitable potential microorganism for use in chitosanase production.
