Optimization of Culture Conditions Using the Method of Response Surface Methodology for Chitosanase Production by Enterobacter sp. YT21

  • Optimization of culture conditions were conducted for chitosanase production by Enterobacter sp. YT21. Firstly Culture temperature, pH and inoculum volume were optimized by one-factor-at-a-time experiments, and then Box-Behnken design and response surface methodology were employed for further optimization of the chitosanase production. The results indicated that the optimal culture conditions were 31.74℃, pH 6.5 and 7.26% (V/V) inoculum, and the highest chitosanase activity obtained was 13.59 U/mL. Error of the actual value compared with the predicted one was only 0.43%, so Enterobacter sp. YT21 was proved to be a suitable potential microorganism for use in chitosanase production.
